  Thissingleenzymeiscomposedoftwosubunits(D1andD12)andhasthreeenzymaticactivities(RNAtriphosphataseandguanylyltransferasebytheD1subunitandguaninemethyltransferasebytheD12subunit);allnecessaryforadditionofacompleteCap0structure,m7Gppp5′N(5,6).Invitrotranscriptscanbecappedinlessthanonehourinthepresenceofthecappingenzyme,reactionbuffer,GTP,andthemethyldonor,SAM.Cappingisnearly100%efficientandallcappedstructuresareaddedintheproperorientation,unlikeco-transcriptionaladditionofsomecapanalogs(7).
